Sky Sports commentator Matthew Macklin, a former boxer, admitted that he'd never seen anything like it before.
Smith was making his debut as a light-heavyweight fighter and was expected to defeat Castillo in the undercard fight to Anthony Joshua's loss to Oleksandr Uzyk.
He delivered with a crushing right-hand that instantly ended the contest.
The North London crowd booed when Castillo became clearly unwell on the canvas.
Referee Bob Williams quickly moved Castillo to the recovery position.
The 33-year old Dominican Republic boxer was then taken to the hospital by ambulance.

Although Smith's win was not as important, it was a great performance by the Liverpool fighter coming off his first defeat against Canelo alvarez.
Smith was accompanied by Buddy McGirt, his new trainer. Smith drove his long levers into McGirt a few times in the opener. He showed no signs that he had sustained any permanent damage from the beating Canelo dealt to his lead arm.
Smith, 31, was able to connect with his victim with a powerful right hand and left him in a very worrying place.
